This is a grant-supported research scientist position supervised by Dr. Jeffrey Carroll. Research in the Carroll lab is primarily focused on enhancing treatments for people impacted by rare neurodegenerative diseases, including Huntington’s disease. The Carroll lab relies on cell and mouse model systems in an attempt to help understand the basic pathological mechanisms of disease, as well as test experimental therapeutics.
In this position, the Research Scientist will assist in conducting grant-sponsored research into the consequences of Huntingtin lowering in mouse and cell models of Huntington’s Disease. They will conduct hands-on research while also playing an important role in project management, data analysis, and drafting manuscripts describing our findings.
DUTIES AND RESPONSIBILITIES
The Research Scientist/Engineer 3 (E S UAW RSE) will have the following responsibilities:
Mouse Work (40%)
  • Assisting in maintaining the mouse colony
  • Sacrifice via intraperitoneal or subcutaneous injection of anesthetics and preparation of plasma samples
  • Dissection of tissues, including micro-dissection of brain tissue into sub-regions
  • Isolation of biomolecules (DNA, RNA and Protein) from tissue and cell samples
  • Treatment of mice via intraperitoneal, intravenous and subcutaneous injections
  • Injection of oliogonucleotide drugs intracerebroventricularly
  • Transcardial perfusion of mice with PBS and fixatives to generated fixed mouse tissues for histological analysis
    Cell and Molecular Biology (40%)
  • Isolation of biomolecules (DNA, RNA and Protein) from tissue and cell samples
  • Quantification of target gene expression with quantitative real time PCR (QRT-PCR) or related techniques
  • Quantification of target protein expression with western blots
  • Staining of tissue slides for immunohistochemistry
  • Analysis of immunohistochemical images using existing protocols in the ImageJ software package
  • Significant experience with cloning plasmid DNA, including propagation of plasmids in E. coli, sub-cloning
  • Generation and validation of guide RNAs for CRISPR/Cas9 experiments
  • Sub-cloning guide-RNA and other cassettes into AAV-generation plasmids
  • Gene expression using RNASequencing techniques
  • Assays in immortalized cell culture including post-stessor viability, sub-cellular trafficking and enzyme activation assays.
    Analyzing Results and Writing Manuscripts (20%)
  • Reads and understands the literature around the topics of research
  • Generates research outputs in the forms of slide decks, manuscript text and figures, progress reports to funding agencies, conference posters and presentations
  • Careful record keeping using Microsoft Office and Cloud Based online Storage
  • Statistical analysis of data using R
  • Analysis of microscopy images using ImageJ and other programs
    MINIMUM REQUIREMENTS
  • Bachelor's degree in Engineering or Scientific Discipline
  • Four years of relevant experience, OR Equivalent combination of education/experience.
    Equivalent education/experience will substitute for all minimum qualifications except when there are legal requirements, such as a license/certification/registration.
    ADDITIONAL REQUIREMENTS
  • Experience with microscopy (light, immunofluorescence and/or confocal)
  • Significant molecular biology experience
  • Significant cell culture experience
  • Significant experience with rodents in research, preferably in neuroscience research
    DESIRED QUALIFICATIONS
  • Significant experience in analyzing genome-wide transcriptional datasets
  • Experience with the R programming language
  • Mouse Behavioral analyses
  • Recovery surgeries in mice, including stereotaxic injections in rodent models
  • Experience with primary mammalian cell cultures and/or iPSC cultures
  • Experience with live-cell microscopy
  • Graduate degree in related field
